小暮健太のクラスメイトで透き通るような美人である。1年前に妹が殺されており、その犯人を捜している。そのためか男性不信なところがある。序盤に「変態」と罵られたりするが、旧校舎から脱出する為に健太と行動を共にするうちに彼を見直し始め、終盤には次第に笑顔を見せるようになっていく。そして、紆余曲折の末に自分達を旧校舎に閉じ込めた黒幕である遺作が時計塔に潜んでいることを知ると、ついに健太と共に時計塔に潜んでいた遺作と対面した。自分達を罠にかけて得意満面の遺作に「見栄とプライドだけ」等辛辣な言葉で遺作の策略がすでに破綻していることを告げて散々に挑発し、遺作を激怒させ怒りで冷静さを失った遺作の隙をついて時計塔のはしごを下りて旧校舎に戻り、あらかじめ用意していたペンキをはしごにぶちまけた。怒り狂った遺作は足元をよく見ずナイフを手にしたままはしごを駆け下りた為彼女の狙い通りペンキで足を滑らせ、あっけなく転落死してしまった。そして遺作の遺したナイフを使い健太が脱出口を開けたため、旧校舎に残っていた仲間を連れて旧校舎を脱出。健太に感謝の言葉を述べた。その後展開次第では、エンディングで健太と結ばれる。このゲームで最も精神的にもタフで頼りになる人。それは最後赤川里香のしたことをあっさり許して赤川さんに罪はないわと言い切ってしまえることからもわかります。そこが美由紀の人としての本当の強さ。本当に強い女性とは彼女のような人を言うのだと思う！
